What is the formula/syntax I need to wirte so that if a value in column 'S' is equal to "Yes" then make ABC certain colour

Pick any cell in A/B/C, say A2
Add a conditional format, formula is

=$S2="Yes"

Applies to

$A$2

Now you can copy it to B2,C2 and it will stick.
And copy down as required
